Crocin
Crocin inhibits the reuptake of dopamine and norepinephrine, increases BDNF expression, and reduces neuroinflammatory cytokines (TNF-α, IL-6). It crosses the blood-brain barrier and directly modulates the limbic system. Studies show crocin reduces corticosterone secretion by downregulating CRH expression in the hypothalamus.

Crocetin
Crocetin exhibits potent antioxidant activity, scavenging reactive oxygen species (ROS) that damage hippocampal neurons. It inhibits NF-κB signaling, reducing neuroinflammation. Crocetin also promotes neurogenesis in the hippocampus — the brain region most vulnerable to chronic cortisol exposure. Studies show 73% reduction in neuroinflammatory markers.

Safranal
Safranal binds to GABA-A receptors as a positive allosteric modulator — similar mechanism to benzodiazepines but without dependency risk. It also inhibits monoamine oxidase (MAO) activity, prolonging the action of serotonin and dopamine. A single dose of safranal delays the cortisol peak response to acute stress (Pouchieu et al. 2023).

Picrocrocin
Picrocrocin acts as a stabilizing agent for crocin and crocetin during digestion, protecting them from premature degradation. It also exhibits mild inhibitory activity on acetylcholinesterase, supporting cognitive function. Affron®'s standardization to Lepticrosalides® ≥3.5% ensures consistent picrocrocin content across batches.
